Changes in Atlantic cod (Gadus morhua L.) sperm quality during the spawning season ArchiMer

The biology of cod reproduction is well described in the scientific literature. However, sperm biology and spermatozoa management are poorly studied in this species. Because of its recent farming expansion, a better knowledge of cod gametes is becoming especially useful. This work aimed at establishing tools to study sperm biology in cod, and also investigated the existence of changes in cod sperm quality during the spawning period. We showed that sperm concentration could be assessed using spectrophotometry at 260 nm. Sperm motility significantly decreased after a 168-h storage at 4 degrees C. La congélation du sperme et des embryons d'huîtres perlières est un moyen de renforcer la sécurité du potentiel économique que représente la Perliculture en Polynésie Française. Elle permet la conservation, à faible coût, de la biodiversité ex-situ des populations d'huîtres perlières sauvages et des génotypes de sujets présentant un intérêt particulier pour l'industrie de la perle. Un avantage complémentaire de la cryoconservation sera de faciliter l'avancement des programmes d'amélioration génétique. Effects of blue light on the biochemical composition and photosynthetic activity of Isochrysis sp. (T-iso) ArchiMer

In aquaculture, particularly in bivalve hatcheries, the biochemical composition of algal diets has a strong influence on larval and post-larval development. Biochemical composition is known to be related to culture conditions, among which light represents a major source of variation. The effects of blue light on biochemical composition and photosynthetic rate of Isochrysis sp. (T-iso) CCAP 927/14 were assessed in chemostat at a single irradiance (300 μmol photons m−2 s−1) and compared with white light. Microalgae offer a high potential for energetic lipid storage as well as high growth rates. They are therefore considered promising candidates for biofuel production, with the selection of high lipid-producing strains a major objective in projects on the development of this technology. We developed a mutation-selection method aimed at increasing microalgae neutral lipid productivity. Genetic structure at different spatial scales in the pearl oyster (Pinctada margaritifera cumingii) in French Polynesian lagoons: beware of sampling strategy and genetic patchiness ArchiMer

In order to study further the genetic structure of the pearl oyster Pinctada margaritifera in French Polynesia with a special consideration for the sampling scale, we analyzed or re-analyzed sets of data based on nuclear DNA markers obtained at different spatial scales. At a large scale (several 1,000 km), the remote Marquesas Islands were confirmed to be significantly differentiated from Tuamotu-Gambier and Society archipelagos, with a marked difference however for the two main islands that are different from each other. The tropical seabass Lates calcarifer was introduced in Tahiti in 1984 and then was domesticated without other introduction. Three successive generations have been obtained in captivity. Tissue samples of the 38 animaIs which represent the total Tahitian broodstock were preserved in alcohol to be genotyped with four microsatellite markers (Yue et al., 2001). Three of four described markers were successfully transferred to the laboratory of genetics of Tahiti (marker LcaM01, LcaM02, LcaM03), as the revelation technology available locally did not allow the use of LcaM04 in routine. Microalgae have a diversity of industrial applications such as feed, food ingredients, depuration processes and energy. However, microalgal production costs could be substantially improved by controlling nutrient intake. Accordingly, a better understanding of microalgal nitrogen metabolism is essential. Using in silico analysis from transcriptomic data concerning the microalgae Tisochrysis lutea, four genes encoding putative high-affinity nitrate/nitrite transporters (TlNrt2) were identified. Effects of a remarkably high overall lipid Tisochrysis lutea strain (T+) upon gross biochemical composition, fatty acid (FA), sterol and lipid class composition of Crassostrea gigas larvae were evaluated and compared with a normal strain of Tisochrysis lutea (T) and the diatom Chaetoceros neogracile (Cg). In a first experiment, the influence of different single diets (T, T+ and Cg) and a bispecific diet (TCg) was studied, whereas, effects of monospecific diets (T and T+) and bispecific diets (TCg and T+Cg) were evaluated in a second experiment. Afin de mener à bien le programme Maîtrise de la Reproduction chez l’huître perlière Pinctada margaritifera dont la perspective est l’amélioration génétique, il est essentiel de contrôler les croisements au niveau individuel. Ces croisements ne pourront être contrôlés que dans la mesure où les gamètes mâles et femelles sont obtenus séparément. Ceci permettra aussi de valider les travaux en cryoconservation des spermatozoïdes et de faciliter la production de polyploïdes. Observation of the larval distribution of two soleidae: Solea solea (L.) and Microchirus variegatus (Donovan) in the Bay of Biscay ArchiMer

Surveys for larval stages of Dover sole (Solea solea) and Thickback sole (Microchirus variegatus) were carried out in the northern part of the bay of Biscay between 1985 and 1989. The spawning periods of these species overlap but the spawning areas are different since young larvae of M. variegatus are recorded more offshore than those of S. solea. In addition, in spite of a similar behaviour the first species undergo smaller vertical migrations than the second and never reach surface layers. These two observations suggest why Thickhack sole larvae on the contrary of Dover sole larvae never reach coastal areas and achieve whole development in water deeper than 40 m. The life cycle of this species developing thus entirely offshore.
